A clinical case of equine fungal placentitis with reference to hormone profiles and ultrasonography OAK
MURASE, Harutaka; NIWA, Hidekazu; KATAYAMA, Yoshinari; SATO, Fumio; HADA, Tetsuro; NAMBO, Yasuo.
Fungal placentitis is an infectious disease inducing abortion in pregnant mares. In the present report, we describe a field case of abortion caused by fungal placentitis with consecutive examinations. The progesterone level and combined thickness of the uterus and placenta (CTUP) were abnormal before the onset of clinical signs. Additionally, the estradiol level started to change before the appearance of clinical signs. Abnormal serum amyloid A values and an abnormal fetal heart rate were observed after the onset of clinical signs. The present report demonstrates that the progesterone level and CTUP may be adequate as early diagnostic markers of fungal placentitis and bacterial infection. Endocrinological evaluation based on cutoff values or serial...

Anti-Müllerian hormone as an indicator of hemi-castrated unilateral cryptorchid horses OAK
MURASE, Harutaka; SAITO, Shigeaki; AMAYA, Tomohiko; SATO, Fumio; BALL, Barry Allen; NAMBO, Yasuo.
Anti-Müllerian hormone (AMH), a glycoprotein secreted from the fetal testis, is responsible for regression of the Müllerian duct in the male fetus. The aim of this study was to evaluate the usefulness of serum AMH as a biomarker for diagnosis of cryptorchidism in horses. Serum AMH concentrations were measured in intact stallions, hemi-castrated unilateral cryptorchid stallions, and geldings. In addition, expression of AMH was characterized in cryptorchid testes by immunohistochemistry. Serum AMH was detected in intact stallions (n=11, 13.3 ± 1.8 ng/ml) and in hemi-castrated cryptorchid stallions (n=8, 17.6 ± 3.0 ng/ml), but not in geldings (n=6, all data were below the limit of detection). Immunolabeling for AMH was detected in Sertoli cells of undescended...

Effects of a single use of the GnRH analog buserelin on the induction of ovulation and endocrine profiles in heavy draft mares OAK
MIKI, Wataru; ONIYAMA, Hiroyuki; TAKEDA, Naomasa; KIMURA, Yuki; HANEDA, Shingo; MATSUI, Motozumi; TAYA, Kazuyoshi; NAMBO, Yasuo.
We observed structural changes in the follicles and uterus of heavy draft mares during estrus and examined the effect of a single injection of the gonadotropin-releasing hormone analog buserelin on ovulation and endocrine profiles. Twenty-two heavy draft mares were divided into a buserelin-treated group (n=8) and a control group (n=14). Mares were given an intramuscular injection of 40 µg buserelin when they presented signs of estrus to a teaser stallion, had ≥45 mm diameter follicles, and presented decreased uterine edema compared with the previous examination. The follicles and uterus were monitored using transrectal ultrasound imaging and measurement of blood levels of luteinizing hormone (LH), follicle-stimulating hormone (FSH), progesterone, and...

Evaluation of Motion Correction Processing in Equine Bone Scintigraphy by Scheffé’s Method of Paired Comparisons OAK
YAMAGUCHI, Toshiro; ENDO, Yoshiro; NAMBO, Yasuo; SATO, Fumio; SASAKI, Naoki; YAMADA, Kazutaka; 南保, 泰雄; 佐々木, 直樹; 山田, 一孝.
Equine bone scintigraphy is usually performed with horses in standing position under sedation. However, swaying motion often leads to poor-quality images. To examine the usefulness of motion correction (MC) processing, equine bone scintigrams were evaluated using Scheffé’s method of paired comparisons. A significant difference in evaluation scores was detected by analysis of variance (F test, P<0.01). According to all observers, Yardstick analysis scores were higher for images use of MC processing than for those no use of MC processing, for all parts. Overall scores of 5 observers were as follows: without MC 100% acquisition time (AT, lowest), use of MC with 25% AT, MC 50% AT, MC 75% AT and MC 100% AT (highest). Thus, MC processing shortens AT in equine...

Experimental investigation of bone mineral density in Thoroughbreds using quantitative computed tomography OAK
YAMADA, Kazutaka; SATO, Fumio; HIGUCHI, Tohru; NISHIHARA, Kaori; KAYANO, Mitsunori; SASAKI, Naoki; NAMBO, Yasuo.
Bone mineral density (BMD) is one of the indications of the strength and health. BMD measured by quantitative computed tomography (QCT) was compared with that measured by dual energy X-ray absorptiometry (DXA) and radiographic bone aluminum equivalence (RBAE). Limbs were removed from horses that had been euthanized for reasons not associated with this study. Sixteen limbs (left and right metacarpals and metatarsals) from 4 horses were used to compare BMD as measured by QCT with those measured by DXA and RBAE. There was a strong correlation between BMD values measured by QCT and those measured by DXA (R2=0.85); correlation was also observed between values obtained by QCT and those obtained by RBAE (R2=0.61). To investigate changes in BMD with age, 37 right...

Hematological and biochemical features of postpartum fever in the heavy draft mare OAK
AOKI, Takahiro; KIMURA, Yuki; OYA, Anna; CHIBA, Akiko; ISHII, Mitsuo; NAMBO, Yasuo.
Heavy draft mares potentially have a higher risk of suffering from postpartum fever (PF) than light breed mares. The purpose of this study was to compare hematological and biochemical features between clinically healthy mares (n=40) and PF-affected mares (n=16). Mares were classified as having PF when their rectal temperature rose to >38.5°C within 96 hr of foaling. The numbers of leukocytes, lymphocytes, and neutrophils and the serum magnesium level were significantly lower in PF-affected mares. The serum SAA and NEFA levels were significantly higher in PF-affected mares. Strong inflammation responses, fat mobilization associated with negative energy balance and possibly endotoxin participate in hematological and biochemical features of PF in heavy...

Relationship between the Placental Retention Time and the Reproductive Performance at the Foal Heat in Thoroughbred and a Comparison with Heavy Draft OAK
ISHII, Mitsuo; AOKI, Takahiro; YAMAKAWA, Kazuhiro; MAGATA, Fumie; GOJO, Chikara; ITO, Katsumi; KAYANO, Mitsunori; NAMBO, Yasuo; 石井, 三都夫.
The aim of this study was to clarify the relationship between the placental retention time (PRT) and the reproductive performance following mating at the foal heat in Thoroughbreds. For this purpose, we interviewed 292 farmers over a period of 3 years with questionnaires evaluating foaling, expulsion of placenta and reproductive performance at the foal heat in 1,432 mares. The obtained data were later compared with a previous study of heavy draft mares. The average of the PRT of the 1,432 Thoroughbred mares was 58 ± 88 min (mean ± SD). The mean PRT of Thoroughbreds was significantly shorter than that of the148 min of heavy draft mares. The incidences of retained placenta (RP) occurring in the Thoroughbred mares were 5.2 and 4.0%, for over 3 and 4 hr after...

Seasonal changes in serum progesterone levels in Thoroughbred racehorses in training OAK
TAKAHASHI, Yuji; AKAI, Makoto; MURASE, Harutaka; NAMBO, Yasuo.
The objective of the present study was to verify the seasonal luteal activity of racehorses in training in Japan from March to August. We allocated 102 horses into a luteal activity group and non-luteal activity group. The luteal activity group included horses with serum progesterone levels that were consistently >1 ng/ml and changed by ± 1 ng/ml. In contrast, the progesterone levels of the non-luteal activity group were consistently <1 ng/ml. In late spring (from May 1 to June 30) and summer (from July 1 to August 31), the percentage of horses in the luteal activity group was significantly higher than in early spring (from March 1 to April 30, P<0.01). These findings demonstrate clear seasonal variations in ovarian activity. The present study...

A total of 87 Thoroughbred horses and 10 ixodid ticks from a ranch in Hidaka district, Hokkaido were tested for tick-borne diseases. Using the indirect fluorescent antibody (IFA) method, 3.4, 92.0 and 97.7% of the horses showed antibody titers of ≥80 against Anaplasma phagocytophilum, Rickettsia helvetica, and Borrelia garinii, respectively. This is the first report of infection with the 3 pathogens in horses in Japan. Using PCR, DNAs from the peripheral blood of all horses were found negative with any Anaplasma, Rickettsia and Borrelia spp., while those from Haemaphysalis megaspinosa ticks were found positive for Anaplasma sp. closely related to A. phagocytophilum in Japan, and A. bovis. B. japonica was also detected in an H. flava tick for the first time.
